Amyloid Beta is a peptide derived from the amyloid precursor protein (APP), which is cleaved by beta secretase and gamma secretase to yield Aβ. Aβ is the main component of Amyloid plaques, which are insoluble clumps of beta proteins that are potentially toxic to neurons and their synapses.
Research suggests that the plaques disrupt communication between brain cells, which would result in cognitive decline.
The Tau protein is a microtubule-associated protein which stabilizes the microtubules that transport intracellular substances in nerve cells. When Tau malfunctions, it detaches from these microtubules and sticks to other Tau molecules, where threads are formed. These threads eventually result in neurofibrillary tangle.
Since Tau is interfering with intraneuronal transport and metabolism during this process, research suggests this plays another major role in cognitive decline.
