mLST8 (mammalian lethal with sec-13), an about 39 kDa protein which has WD40 domains, has been identified as a configuration factor of mTORC1/mTOR complex 1 and then found to be also a configuration factor of mTORC2. mLST8 is conserved over species and widely expressed in a variety of cells. It is assumed that mLST8 plays a role in promoting activity of mTOR kinase, for mLST8 binds to the kinase domain of mTOR and phosphorylation of p70 S6 (a substrate of mTOR kinase) decreases by RNAi.
