Rheb (ras homolog enriched in brain) is a 20 kDa protein identified as a Ras homolog expressed in the rat hippocampus, but it widely exists in a variety of cells. It is also reported that GTP-binding form is predominant in Rheb, unlike other low molecular weight GTP-binding proteins. While it is clear that GAP of Rheb is TSC2 which is the gene product of TS (tuberous sclerosis), TCTP (translationally controlled tumor protein) is reported to be expected as a GEF of Rheb. It is known that over-expression of Rheb can rescue the function of mTOR from inactivation by amino-acid starvation, and it is clear that Rheb acts as an activator of mTOR signaling cascade.
