TEK, also known as Tie-2 (Tyrosine kinase with Ig-like loops and Epidermal growth factor homology domains-2) and CD202b, is a 145 kDa receptor tyrosine kinase expressed on endothelial cells and hematopoietic cells. Its structure possesses an extracellular domain containing two immunoglobulin-like loops separated by three epidermal growth factor-like repeats that are connected to three fibronectin type lll-like repeats, and it has a membrane-penetrating domain and two tyrosine kinase domains in the intracellular region. As ligands, Angiopoietin (Ang)-1 and -4 induce phosphorylation of the receptor while Ang-2 and -3 appear to act as antagonists for its activation. This signaling pathway can regulate cellular adhesion to vascular endothelium and could be important for maintaining endothelial cell survival and integrity in adult blood vessels as well as tumor angiogenesis. This monoclonal antibody recognizes TEK/Tie-2 by W.B. and I.P. applications and it enables FACS analysis of TEK/Tie-2-expressing cells. It has been confirmed that the addition of this antibody does not induce ligand-binding-dependent phosphorylation of TEK/Tie-2 and it may be used as a neutralizing antibody.
