Radioimmunoassay (RIA) for the determination Vasopressin in plasma or urine.<br><br>
After solid phase extraction (SPE) or ethanol extraction of the plasma, vasopressin is assayed by a competitive radioimmunoassay. Urinary vasopressin can be measured directly. This assay uses a rabbit anti-vasopressin antiserum and a radioiodinated vasopressin [125I] tracer. Bound and free phases are separated by a second antibody bound to solid phase particles, followed by a centrifugation step. The radioactivity in the bound fractions is measured and a typical calibration curve can be generated. The values of the extracted unknowns are corrected for extraction recovery. Also known as Arginine Vasopressin (AVP), vasopressin is a cyclic nanopeptide with a molecular weight of 1083 Daltons. Most of it is stored in the posterior pituitary to be released into the blood stream; However, a part of it is also released directly into the brain.<br><br>
One of the most important roles of Vasopressin is to regulate the body's retention of water: when the body is dehydrated. Vasopressin is released causing the kidneys to conserve water, thus concentrating the urine, and reducing urine volume. It is also involved in the blood circulation because it increases the resistance of the peripheral vessels and thus increases arterial blood pressure. Vasopressin released within the brain has many actions. It is suggested that AVP has implicated memory formation, including: delayed reflexes, image, short- and long-term memory.
